ЧЕТЫРЕХФОТОННАЯ ЛАЗЕРНАЯ СПЕКТРОСКОПИЯ ГИДРАТНЫХ ОБОЛОЧЕК БИОМОЛЕКУЛ И НАНОЧАСТИЦ В ВОДНЫХ РАСТВОРАХ

Спектроскопия четырехфотонного рассеяния (ЧФР) была применена для регистрации вращательных резонансов молекул H2O и H2O2 в водных растворах белков, ДНК, денатурированной ДНК и углеродных нанотрубок в диапазоне ±10 cм-1 со спектральным разрешением 3 ГГц. В этих спектрах зарегистрированы резонансы вращательного спектра молекулы Н2О2, а также  орто- и пара-изомеров молекулы Н2О. Обнаружен значительный рост резонансного вклада вращательных переходов этих молекул в растворе по сравнению с дистиллированной водой. Этот факт может быть интерпретирован как проявление специфических свойств гидратного слоя на границе воды с биомолекулами и углеродными нанотрубками. Анализ спектров ЧФР показал, что концентрация молекул H2O2 в гидратном слое молекулы ДНК выросла в 3 раза после денатурации. Были получены спектры ЧФР водных растворов белка a-химотрипсин в спектральном диапазоне ± 7 cм-1 и концентрациях от 0 до 20 мг/cм3. Обнаружено, что скорость гиперзвука водного раствора белка, измеряемая по сдвигу компонент  Мандельштама-Бриллюэна (МБ) в спектре рассеяния, имеет кубическую зависимость от концентрации и достигает 3000 м/с при 20 мг/cм3.

The considerable growth of H2O and H2O2 rotational lines intensity in FP spectra of biopolymer aqueous solutions have been observed. The rotational lines of ortho spin isomer H2O increases much stronger than para isomer’s lines.

The low frequency modes of biopolymer aqueous solutions carry the unique information on the structure of these bio molecules and its interactions with solvent. Unfortunately, usual Raman or THZ spectroscopy in the range 0.1-10 cm-1 doesn’t operate successfully in aqueous solutions. We have developed four-photon (FP) techniques based on visible pump and Stokes laser beams whose frequency difference (w w S) is scanned in the microwave range (3-3000 GHz) and can be tuned to the resonance with any vibrational or rotational transition of solute or solvent in the range 0.1-100 cm-1. We have observed the considerable growth (by factor of 10) of H2O and H2O2 molecules rotational lines intensity in FP spectra due to the presence of biopolymers (protein, DNA, denatured DNA) in aqueous solutions. This reflects the increasing of quasi-free H2O molecules concentration and hydrogen peroxide production in water/biopolymer hydration shell. Four-photon spectra fitting shows the enhancement of the H2O2 rotational line’s amplitude by a factor of ~3 in denatured DNA aqueous solution, when the denaturizing process has been completed.

We have found that the rotational lines intensity of H2O molecule ortho isomer increases in the biopolymer’s aqueous solutions much stronger than the para isomer’s lines. One can suggest that the biological macromolecules interact selectively with H2O spin isomers at the nearest surrounding layers.

It is well known that biological molecule hydration leads to ice-like structurization of water molecules ensemble in the hydration shell of the solute. This fact is qualitatively confirmed by the growth of Brillouin shift and solution’s stiffness upon protein concentration increasing in a-chymotrypsin aqueous solution. Similar sound velocity growth we have observed in DNA solution after denaturation (Fig.1). Bold arrows in Fig.1 mark the position of Brillouin resonances of DNA (1) or denatured DNA (3) aqueous solutions (Fig.1).
